Influence of compression method on physical properties and release rate of verapamil hydrochloride. Pellets are spherical granules (0,5-2,0 mm) used in hard gelatin capsules or compressed into tablets. It is an example of multiparticulate drug delivery. Compression of pellets into tablets is a modern technological process, is much more perfect than enclosing them in a hard gelatin capsule. The aim of this study was to evaluate the influence of a compression method of floating pellets on physical properties of tablets and release rate of verapamil hydrochloride (VH). The assumption of the study was to get release profile VH from tablets similar to pellets before compression. For investigation single stroke tablet press Korsch EK0 and high tech rotary tablet press ERWEKA RTP-D8 connected with PC were used. Pellets were prepared by extrusion and spheronization method, and coated with polymer film - Eudragit NE. For tablet masses as excipients Avicel PH 101, Tablettose 80 and Sorbitol were used. In all cases of tablets compressed by single stroke tablet press increase of release rate was observed. It was caused probably by film damage under pressing force. The film damage may also have occurred as a result of core deformation. In tablets prepared by rotary tablet press, with increasing the pressure force, a decrease release rate was observed. The release profile from tablets prepared by rotary tablet press was most similar to profile from the pellet before compression. It was caused by equal application of the main and initial pressure force by upper and lower punch.
