Application of thin-layer chromatography to identify spironolactone in selected pharmaceutical formulations

Spironolactone is a synthetic steroid of diuretics group used in treatment of arterial hypertension and in oedemas connected with circulatory insu ciency in form of selected pharmaceuticals. One of the methods applied in determination of spironolactone identity and recommended by Polish Farmacopoeia is thin-layer chromatography in normal phase (NP-TLC). In this work, the optimal chromatographic conditions such as: kind of stationary phase, volume composition of mobile phase and visualization of spironolactone spots as bioactive substance in selected commercial preparations using TLC method in normal and reversed system (NP-TLC and RP-TLC) were determined. On the basis of obtained results it was concluded that, the developed TLC method is e ective and can be used in qualitative analysis of pharmaceutical preparations containing spironolactone as bioactive substance.
