Capillary electrophoresis as a modern tool for chiral analysis of medicinal products. Part I. Enantioselection mechanism in capillary electrophoresis, chiral selectors. The separation of enantiomers is a challenge in the pharmacy, but also in the chemistry and biology. The stereochemistry of enantiomers can affect their biological activities, therefore developing of a method for their purification and separation is of great interest. Capillary electrophoresis (CE) is a modern and advantageous tool in the chiral separation technique. This technique provides a number of advantages in chiral separations when compared with other methods: a high separation efficiency as well as flexibility in relation to optimization of chiral separation.
The separation of chiral compounds can be achieved in CE by simple addition to the background electrolyte of an appropriate chiral selector, e.g., native and derivatized cyclodextrin, natural and synthetic chiral micelles, crown ethers, chiral ligands, protein, oligo- and polysaccharides, and macrocyclic antibiotics. Among different chiral selectors, cyclodextrins and their derivatives are the most widely employed in chiral CE techniques. In the first part of this article cyclodextrin and crown ethers are described. The resolution mechanism, when CDs and crown ethers are used as chiral selectors, is usually based on inclusion complexation.
