Influence of disintegrants on teophylline release rate. In the first step of the study disintegration of empty capsules made of gelatine, hypromelose or polyvinylalcohol/starch blend was compared and for further experiments gelatine capsules were used as their disintegration time was less than 8 min. The aim of this study was to evaluate the effect of combinations of two disintegrants on the release of theophylline (100 mg) from capsules in a basket apparatus at 50 rpm. Modified corn starch (PharmGel), pregelatinised starch (StarCap 1500), sodium starch glycolate (Primojel), croscarmellose sodium (Ac-Di-Sol) and alginic acid (Protacid) were used as disintegrating agents. Out of the 24 formulations only one, based on croscarmellose sodium (15%) and modified corn starch (25%) ensured release of more than 80% of theophylline in 20 min. Although combinations of these two disintegrants may be indicated as generelly most efficient, but no simple relationship between disintegrants type and their concentration and proportion is found.
