Antileukorienes - benefit and risk. Leukotrienes are the products of the 5-LOX pathway of arachidonic acid metabolism. Their diverse and potent actions are associated with specific receptors. Leukotrienes are key mediators in asthma, allergic rhinitis and atopic dermatitis. They are also considered to be central to the pathogenesis of cystic fibrosis, atherosclerosis and cancer. Antileukotrienes are a new class of anti-inflammatory drugs that interfere directly with leukotriene production (zileuton) or receptors (montelukast, zafirlukast, pranlukast). This article reviews their pharmacology, pharmacokinetics, clinical efficacy and adverse effects.
