Rola partenolidu w skojarzonej terapii przeciwnowotworowej

Partenolid jako lakton seskwiterpenowy złocienia maruny (Tanacetum parthenium) oprócz właściwości przeciwmigrenowych i przeciwzapalnych wykazuje również właściwości przeciwnowotworowe. Hamuje on aktywność czynników transkrypcyjnych NF-kB i STAT oraz indukuje trwałą aktywację JNK i p53, skutkiem czego może uwrażliwiać komórki nowotworowe na radio- i chemioterapię. Partenolid w komórkach nowotworowych redukuje poziom glutationu i doprowadza do kumulacji uszkodzeń wywołanych przez reaktywne formy tlenu. Przyczynia się tym samym do indukcji stresu oksydacyjnego i apoptozy. Pomimo wielu zaawansowanych badań dotyczących nowych leków, okazuje się, że skuteczność terapii przeciwnowotworowych jest ograniczona najczęściej wysokim stopniem heterogenności nowotworu lub złożonością dróg sygnalizacji komórkowej. Same postępy wynikające z doskonalenia metod terapeutycznych są niewystarczające. Aby poprawić wyniki samej terapii, stosuje się koncepcję kojarzenia metod. W ostatniej dekadzie pojawiło się wiele doniesień na temat roli partenolidu w hamowaniu wzrostu nowotworu in vitro. Poniższy artykuł stanowi podsumowanie badań dotyczących skuteczności chemioterapii poprzez terapię skojarzeniową z partenolidem.

The role of parthenolide in combination anti-cancer therapy

Parthenolide – sesquiterpene lactone (Tanacetum parthenium) is known, primarily, for its anti-inflammatory and anti‑migraine activities. It has been postulated that their anti-tumour activity depends on transcription factor NF-kB, STAT inhibition and induces lasting activation of JNK and p53. Due to above the „effect” of cells sensitization to radio- and chemo-therapy can be observed. Parthenolide, in tumour cells, reduced glutathione level and led to reactive oxygen species accumulation. Many advanced studies, on the new drugs fields, have shown that the effectiveness of cancer treatments is usually limited by a high degree of heterogeneity of the tumour or the complexity of cellular signalling routes. Therefore the progress of new therapeutic methods and the improvement of classic ones are inadequate. Due to that, the attention should be placed on the concept of combination treatment with different methods/strategies. In the last decade many reports on the inhibition role of isolated parthenolide on tumour growth in vitro have been published. Additionally same research groups published the therapeutic effects of the parthenolide in combination with other drugs used in anticancer therapy. These review article try to summarise the state of current researches on the effectiveness of chemo/radio therapy by combining therapy with parthenolide.
