Wpływ doustnych leków hipoglikemizujących na aktywność katepsyny B w granulocytach obojętnochłonnych

Influence of oral hipolipemic drugs on Cathepsin B activity in polymorphonuclear leukocytes. Cathepsin B (CB) is a cysteine protease present in the large amount in lysosomes especially in tissues with intensive metabolism as well as in polymorphonuclear leukocytes (PMN). CB plays crucial role in turn over of different proteins (strange and own) and participates also in host defense. In diabetic patients functions of PMN are disturbed and what follows their proteinases activities also. The medicines applied in diabetes have the multidirectional working (pleiotropic effect) and can modulate also the PMN function as well as cathepsin B. PMN isolated from blood of healthy people and patients with type 2 diabetes mellitus were incubated first of all with solution of metformin and gliclizide (25mM) by three hours and next step in three different concentrations (25; 12,5 i 5mM) by one hours. The results of these investigations can not unambiguously define which medicine was more effective on the CB activities but the most significant changes, percentage increase of activity in diabetic patients, have shown metformin.
